An optimized, simplified workflow for the targeted proteomic analysis of fibrinogen in 4,200 plasma samples

Presenter Bio: Dr. Stefani Thomas is an Assistant Professor in the Department of Laboratory Medicine and Pathology at the University of Minnesota, and the Associate Medical Director of the M Health Fairview University of Minnesota Medical Center West Bank Laboratory. She earned a BA in Biological Sciences from Dartmouth College, a PhD in Pharmaceutical Sciences from the University of Southern California, and she completed a Clinical Chemistry postdoctoral fellowship at Johns Hopkins. Her research program at the University of Minnesota utilizes mass spectrometry-based clinical proteomics for therapeutic and diagnostic biomarker development.


Summary

The plasma concentration of fibrinogen – the major structural component of blood clots – is a critical indicator of hemostasis. Although plasma fibrinogen immunoassays performed in clinical laboratories generally have high accuracy and sensitivity, they require a large sample volume and they are relatively expensive. To avoid these limitations for the analysis of plasma fibrinogen in a longitudinal clinical research study (n=4,200 participants), we developed and validated a targeted mass spectrometry assay using a Thermo Scientific™ Q Exactive™ Plus mass spectrometer coupled with a Thermo Scientific™ Vanquish™ UHPLC system, and we streamlined the sample processing workflow to successfully achieve a throughput of ~90 samples/24 hours.
